Select Sites & Features > Restriction Fragment Summary to view a tabular summary for Enzymes - Restriction Map methods applied to the assay surface.
• The restriction enzymes selected from the assay surface will appear in the tabular summary. If none are selected, then all will appear in the tabular summary.
• The summary lists the following data for single, double, and triple digests; the number and position of cuts; the enzyme responsible for each cut; and the length of the fragment.
The following options are used in conjunction with Enzymes - Restriction Map methods.
• Linearize Sequence: Select Sites & Features > Linearize Sequence to reverse GeneQuest’s assumption that a given sequence is circular. This is especially important in restriction site analysis, since GeneQuest may find fewer cut sites on a linear strand than on a circular one.
• Sub-range Context: Select Sites & Features > Sub-range Context to search for restriction sites in the sequence where the recognition site lies outside the sub-range of sequence specified.
Note: Sub-range Context has no effect unless you have used the Set Ends button.
• Include Uncertain Sites: Select Sites & Features > Include Uncertain to label potential restriction sites by favorably interpreting ambiguous DNA residues. For example, the sequence ANGCTT, an uncertain site for HindIII (AAGCTT), would be labeled as a cut site.
